Visceral tissues of oyster were collected in this study. A primer set derived from Heat shock protein sequence was used to amplify fragment from oyster. And the PCR products was cloned, screen and transformed into E. coli. These bacteria were cultured and immune rabbit to collect anti-hrp serum. Following serum antibody titer test, ELISA, immune blot test to available of heat shock proteins within oysters. Result showed that the cloned fragment has the identical sequence with oyster heat-shock protein 90. The transformed sequence was prepared using 1 ml of inactive bacilli with the same volume of adjuvant to immune rabbits. After immunization, serum antibody titer was test by ELISA and immune blot test to demonstrate that their ability to neutralize heat shock protein 90. Artificial experiment by preserving oysters at 37 ℃?for 5 minutes and muscle, gonad and hepatopancreas were collected and examined their ability to response to temperature stress. Results revealed that the concentration of the heat-shock protein increased and have shown variable response in different organs. For the time being, stress factors consisted of changing of salinity and temperature were performed to study the response and ability of heat shock proteins in oyster. This study has shown its advantage in clinical application, and future applied in the field will be done. |
