In the aquaculture pond and shipping system, we frequently encounter the stress conditions for fish, such as abnormal water temperature, low dissolved oxygen, improper salinity, high density, infectious disease and other adverse living environment. Responding to these stress environments, fish will drive some of specific genes to increase specific proteins in order to survive. Most studies emphasize the expression patterns of some specific genes to face a particular stress. However, it is hardly to find out a superior brookstock which has special genetic inheritances to survive under the adversity in the culture environment. Therefore, in this one-year project, we will plan to use Africa cichlid, such as Libidochromis caeruleus or Pseudotropheus zebra, as experimental animals simply because they are highly economic exported ornamental fish in Taiwan and they are short generation time so that it is possible to generate a genetic stock within a year. We will screen out many individuals which enable to survive under the high or low temperature stress. We will confirm whether this superior genetic trait can be to next generations. The putative founders G0 among examined fish will be selected and cultured after they mate with other individual and their superior traits can be passed to next generations. Next, we will identify this special superior brookstock family by using various microsatellite analysis of its genomic DNA. When the screen platform of high-quality varieties has been established, it is possible for us to achieve the first step goal that is we develop and identify the superior variety which cans appropriately response to the high or low temperature without detriment to environmental stress. |
