In this one-year proposal, we have developed a gene transfer platform that works efficiently on African killifish, one of freshwater ornamental fishes. Heterogeneous genes coding for green fluorescent protein was transferred into the one-celled stage of African killifish. In total, 997 fertilized eggs were transferred. The survival rate of transferred embryos was 82%, which was similar to that of the control group. After the transferred embryos are cultured, we found that 114 embryos (around 14% of original injected eggs) appearing mosaic green fluorescence. We selected 38 embryos that displayed stronger fluorescent signal and cultured to adulthood. After crossing with the wild-type individuals, there was one line at 7 days-postfertilization exhibited GFP throughout the whole body not only under the fluorescent microscope but also under the normal light source. We believe that using molecular biotechnological approach can enhance the color patterns of killifish, resulting in increasing the commercial value of this killifish species that has dull body color and boring pattern. |
