The full length of interleukin-1β (IL-1β) was cloned from grouper by RACE (rapid amplification of cDNA ends). IL-1β protein was expressed in E. coli and was further purified then confirmed by SDS-PAGE. White blood cell (WBC) culture from head kidney demonstrated that IL-1β could promote superoxide anion strength in both black porgy and grouper. IL-1β could also block the inhibitory effect caused by dexamethasone on WBC superoxide anion strength. Grouper infected with E. coli, exhibited a significant increase in IL-1β gene expression in head kidney and spleen. Infecting grouper with Vibrio anguillarum, followed by treated with recombinant IL-1β, showed that IL-1β could promote the WBC superoxide anion strength in WBC cell culture from head kidney. Thus, the above results suggest that recombinant IL-1β promotes immune response in marine fish (black porgy and grouper). |
