Research and development of a rapid, sensitive and automatic detection platform is one of the major approaches of controlling viral diseases in aquaculture. In this project, aiming at grouper nervous necrosis virus (NNV), attempts were made to develop an ultra-high sensitive MagnetoReduction Assay (MRA) for quantitively detect the target bio-molecules (NNV viral antigens). Viral encephalopathy and retinopathy (VER), caused by betanodaviruses (piscine nodavirus), is one of the most devastating, occurring in a variety of marine fish all over the world. Firstly, basically using a solution having homogeneously dispersed magnetic nanoparticles, which are coated with anti-NNV antibody. Under external multiple ac magnetic fields, magnetic nanoparticles oscillate with the multiple ac magnetic fields via magnetic interaction. In the premilanary results, we already establish the standard curve in different NNV concentration. The results showed the sensitivity reaches 102 TCID50ml-1 virus titer. We also using NNV reagent to test the IPNV and iridovirus. The reading values were below the backgroup noise. The results suggested the platform specificity is reliable. |
